چکیده

Most of the literature dealing with the participation of citrovorum factor and vitamin Blz in the formation and transfer of l-carbon intermediates stresses the donor compounds serine, glycine, and formate as precursors of the 5-methyl group of thymidine (2). Uracil deoxyriboside, first isolated in crystalline form by Dekker and Todd (3) in 1950, is known to arise as a result of the deamination of cytosine at the structural level of polynucleotide or nucleoside (4-7). Uracil deoxyriboside can also be directly synthesized by the enzymatic attachment of the deoxyribosyl group to uracil (5,8-10). In this paper, evidence is presented that uracil deoxyriboside, which structurally is the simple demethylated form of thymidine, may indeed be the primary acceptor of the methyl group.

systems indicate that the type of halogen, fluorine, bro mine, iodine, or chlorine, substituted at the 5-position of uracil deoxyriboside greatly influences the activity and the type of inhibition of cell proliferation. 5-Fluorodeoxy uridine (FUdR) inhibits deoxyribonucleic acid (DNA) synthesis by blocking the methylation of deoxyuridylic acid to form thymidylic acid (10, 19, 20), whereas bromo...

Feedback Inhibition of Thymidine Kinase by Thymidine Triphosphate.

That the regulation of cell division and deoxyribonucleic acid synthesis is subject to some kind of chemical control in higher organisms is widely accepted (2-5). Davidson (2) has suggested that deoxyribonucleic acid synthesis takes place at critical concentrations of the deoxyribonucleoside triphosphates and that these compounds are components of a homeostatic mechanism. There are now at least...

Incorporation of uracil-2-C14 into the nucleic acids of Neurospora crassa.

The pyrimidine-deficient mutant (1298) of Neurospora crassa, utilizes erotic acid, uracil, uridine, or cytidine for growth, but neither cytosine nor thymine (1). It seems likely, therefore, that this mutant can convert erotic acid and uracil to the pyrimidine bases of ribonucleic acid and deoxyribonucleic acid, and can utilize the nucleoside directly for ribonucleic acid biosynthesis and for co...

Synthesis of pyrimidin-2-one deoxyribosides and their ability to support the growth of the deoxyriboside-requiring organism Lactobacillus acidophilus R 26.

In the past, several methods for reducing pyrimidine nucleosides and nucleotides have been described. Levene & LaForge (1912) reduced uridine and cytidine with hydrogen by using palladium as catalyst. Burke (1955) made use of sodium and ethanol in liquid ammonia. Also, hydrogen with rhodium on alumina (Cohn & Doherty, 1956; Green & Cohen, 1957) was shown to reduce specifically the double bond i...
